salmonella identification from foods in eight hours: a prototype study with salmonella typhimurium.

چکیده
background and objectives: the significant rise in food borne infections is mainly caused by campylobacter spp., salmonella serovars and verotoxigenic escherichia coli . as the emerging food borne pathogens cause disease, more studies have been conducted for rapid detection of these pathogens. the combination of immunomagnetic separation and polymerase chain reaction (ims-pcr) is the most accurate and rapid test preferred by almost every researcher. fourier transform infrared spectroscopy (ftir) is preferred for being a new, user friendly and rapid technique in microbiological analyses. the main aim of this study is to detect application of ims-ftir for salmonella identification from foods in a short time with a higher sensitivity. materials and methods: conventional culture technique (cc), ims-cc, ims-pcr and ims-ftir techniques were compared with each other for rapid detection in artificially contaminated minced beef with salmonella typhimurium, as of the 2nd, 4th and 8th hours of contamination. the method was evaluated in different food matrices and sensitivity, specifity and overall recovery was calculated. results: the results indicate that ims-ftir can detect s. typhimurium as of the 8th hour with sensitivity of 95.6667, accuracy of 91.69329, false positive ratio of 0.04333 and overall recovery of 95.66%. conclusion: it can be suggested that the ims-ftir method is capable of detecting s .typhimurium in a short time with lower cost.

متن کاملIdentification of Salmonella spp. and Salmonella typhimurium by a multiplex PCR-based assay from poultry carcasses in Mashhad- Iran
Poultry meat has been identified as one of the principal foodborne sources of Salmonella. In this preliminary study the prevalence of Salmonella spp. and its typhimurium serovar contamination of broiler carcasses, were determined. Using the rinse test method, numbers of 60 samples, representing 20 broiler flocks, were collected from poultry carcasses after the chilling stage in the processing l...
متن کاملMolecular Characterization of a Salmonella Typhimurium Isolate from Caspian Pony
Typhoid disease or salmonellosis is a common sickness in horses. In several epidemiological studies inhospitalized horses, several serotypes of Salmonella often are predominant in nosocomial infections.Transportation, overcrowding, dehydration, oral antimicrobial therapy and infections are the risk factorswhich may activate latent or subclinical salmonellosis. In this study, t...
متن کاملCOMPARATIVE CHARACTERIZATION OF PORINS FROM SALMONELLA TYPHIO-901 AND SALMONELLA TYPHIMURIUM RA-30
Porins from Salmonella typhi 0-901 and Salmonella typhimurium Ra-30 were characterized and compared. The elution profile of porins from these salmonella species on Sepharose-48 and HPLC appear to be very similar. The findings were confirmed by the electrophoretic pattern which showed three types of porins, i.e. OmpC, OmpD and OmpF in both species. These porins appear to be similar, if not ...
متن کاملSeparation of Salmonella Typhimurium Bacteria from Water Using MWCNTs Arrays
In this study, Salmonella Typhimurium bacteria removal from polluted water has been investigated using multiwall carbon nanotubes arrays. Experimental results reveal that the contact time, the bacterial concentration and the weight of multiwall carbon nanotubes arrays have positive significant effects on the bacteria removal efficiency. Increasing the contact time and the weight of multiwall ca...
متن کاملThreonine Deaminase from Salmonella typhimurium
Purified threonine deaminase of Salmonella typhimurium was found to be monodisperse and to have a molecular weight of 194,000. The protein, in 6 M guanidine HCI and 0.1 M P-mercaptoethanol, dissociates into polypeptide chains of molecular weight 48,500. Tryptic peptide analysis implies that the four component chains of the native enzyme are of identical amino acid sequence. L-Isoleucine and L-v...
